Retatrutide adds glucagon receptor agonism to the dual incretin backbone — recruiting energy expenditure alongside appetite regulation. In TRIUMPH-1, its phase 3 trial in 2,339 adults with obesity but without diabetes, the 12 mg dose produced 28.3% mean weight loss (70.3 lbs) at 80 weeks — with 45.3% of participants losing at least 30%, a level long associated with bariatric surgery. A two-year extension averaged 30.3%. An FDA filing is expected in Q1 2027.
Lyophilized vials: keep sealed, dry, and away from light and heat. Refrigeration at 2–8°C is the best default; −20°C for long-term storage. After reconstitution: refrigerate at 2–8°C and use promptly — no beyond-use date is validated.
Three receptors, one molecule. The most ambitious incretin in development.
